pH-spectra of the tRNase Z activity. Relative k_obs values for the T.maritima pre-tRNA^Arg(₇₄CCA₇₆) cleavage reaction by T.maritima wild-type tRNase Z and its H222K variant are plotted against pH. The fluorescein-labeled pre-tRNA (20 nM) was reacted with each variant (700 nM) at pH 7–10 at 25°C.
