Radiolabeling of total lipids with [14C]acetate in isolated plastids of cotton embryos (34 DPA) and tobacco cell suspensions and chloroplasts for 1 h
| Treatment | Embryos | Cell Suspensions | Leaves |
|---|---|---|---|
| Controla | 9.8 ± 1.8 | 16.6 ± 2.8 | 4.2 ± 0.5 |
| Ethoxyozlamide | 3.2 ± 0.5 | 8.4 ± 2.1 | 1.8 ± 0.6 |
| Acetazolamide | 4.3 ± 0.9 | 12.2 ± 1.9 | NDb |
Approximately 75 μg of embryo or tobacco plastid proteins or 0.167 mg of tobacco chloroplast chlorophylls were pre-incubated with 10 mm of ethoxyzolamide or acetazolamide for 30 min prior to the addition of [14C]acetate. Values represent the mean and sd of three independent experiments. Rates of lipid synthesis in embryo or tobacco cell plastids are represented as nmol acetate h−1 mg−1 protein and for tobacco chloroplasts as nmol acetate h−1 mg−1 chlorophyll.
Measured with DMSO; without DMSO, rates were 9.8 ± 1.8, 16.6 ± 2.8, and 4.2 ± 0.5 for plastids isolated from embryos, cell suspensions, and leaves, respectively.
