Effects of SSF1,
CNS1, and HCH1 on p60v-src
activity in E381K cells. E381K cells expressing an empty vector or a
vector expressing HSP82, SSF1,
CNS1, or HCH1 were transformed with a
galactose-inducible p60v-src expression vector. Cells grown
to mid-logarithmic phase at 25°C in raffinose-containing medium were
shifted to galactose-containing medium for 6 hours to induce expression
of p60v-src. Total cellular proteins were isolated,
separated on 7.5% SDS/PAGE gels and transferred to Immobilon-P
membranes. (A) Equal sample loading was confirmed by
Coomassie blue staining. (B) p60v-src
activity was assessed by reacting the blot with an antibody specific
for phosphotyrosine. (C) p60v-src accumulation
was assessed by reacting the blot with an antibody specific for
p60v-src.
