Serum-starved COS-7 cells were pretreated with AG1478 (200 nM for 10 min) or vehicle DMSO and stimulated with 100 nM PMA. Cell extracts were separated by SDS/PAGE, and Akt phosphorylation on Ser⁴⁷³ was assayed with a phosphorylation site-specific antibody (upper panel).
