Rafts prepared from infected cells contain infectious viral particles. (A) Quantification of virus-associated particles along the sucrose gradient. Caco-2 cells were infected at 10 PFU/cell and were collected 24 h p.i. Whole-cell homogenates were subjected to a sucrose gradient as described in Materials and Methods. Fractions were collected, and the rotavirus titer in each fraction was determined as previously described (40). The data are representative of three independent experiments. (B) Caco-2 cells were infected at 10 PFU/cell and were collected 24 h p.i. DRM were purified by flotation from these infected cells and used to infect MA 104 cells, which were screened by indirect IF for the presence of rotavirus using an anti-RF antibody and a secondary FITC-labeled anti-rabbit IgG antibody. Nuclei were stained with propidium iodide.
