TABLE 1.

Cholesterol and sphingomyelin content of DRM from Caco-2 cells is not modified by rotavirus infectiona

Lipid Contentb (nmol/mg of protein in homogenate [%]) in:
Infected cells Mock-infected cells
Cholesterol 13.7 ± 1.4 [32] 14.2 ± 1.6 [32]
Sphingomyelin 6.1 ± 1.5 [29] 6.7 ± 1.1 [33]
GPL 20.4 ± 2.2 [10] 21.5 ± 1.9 [11]
a

Flasks of Caco-2 cells (75 cm2) were infected with rotavirus or mock infected. At 18 h p.i. DRM were prepared on a sucrose gradient as for Fig. 3. Lipids from whole-cell homogenates and DRM fractions were extracted with chloroform-methanol as described in Materials and Methods.

Results are means ± standard deviations from three independent experiments. Percentages (in brackets) were derived by dividing the amount of a given lipid in the DRM fraction by the amount of the same lipid in the whole-cell homogenate. Cholesterol was assayed from the lipid extract by a colorimetric assay. Glycerophospholipids (GPL) and sphingomyelin were quantified by phosphorus assay, sphingomyelin being assayed after TLC separation.
