Cells expressing SERT mutant I172C (second column) or membranes prepared from those cells (third column) or cells expressing SERT C109A (fourth column) were treated with MTSET for 10 min. Samples of the treated cells or membranes were further incubated with DTT for 10 min before measuring 5-HT uptake or β-CIT binding. Results are expressed as the percentage of control transport in cells or binding in membranes that had not been treated with any of these reagents. All incubations were in PBS containing 0.1 mM CaCl₂ and 1 mM MgCl₂.
