GM130- or giantin-positive nascent Golgi structures accumulate faster than GalNAcT2- or GPP130-positive structures during early telophase. HeLa cells stably transfected for GalNAcT2-GFP were stained with DAPI to visualize chromosomes and GM130 antibody. Wild-type HeLa cells were separately stained with giantin or GPP130 antibodies. Individual cells in the asynchronous population were staged with respect to mitotic phase by appearance of their chromosomes and cell shape in phase-contrast micrographs. All micrographs were taken with a CARV II spinning disk confocal device. The number of cells scored for each data point varied between 9 and 11. (A1–A4) chromosome appearance; DAPI staining.
