Three-week-old PNP–/– mice were injected i.p. twice weekly with 0.5 U/g body wt of TAT-PNP or nonfused PNP (5–8 mice per group). Serum samples, drawn at the indicated time points during 24 weeks of TAT-PNP treatment (squares) or 12 weeks of treatment with nonfused PNP (triangles) were assessed for the presence of antibodies by ELISA. Serum samples were diluted 1:100 and added for 2 hours to plates coated with TAT-PNP or nonfused PNP (5 μg/ml). Following washes, HRP-labeled anti-mouse IgG was added. The assay was developed using TMB substrate. OD was read at 450 nm. Results are mean ± SD.
