(B) PCR amplification with primers flanking the mutation site, followed by BstU1 restriction digest of PCR products clearly distinguishes between wild-type, homozygous, and heterozygous DNA samples. The wild-type allele yields a 213 bp product, which is not cut by BstU1. The bpk mutant allele yields a 215 bp product, which is cut into 125 bp and 90 bp fragments.
