Effect of M40403, on DNA damage and cellular dysfunction in macrophages taken from carrageenan-challenged rats. When evaluated at 4 h post carrageenan injection, a significant damage of DNA (as measured by DNA single strand breakage) was observed in pleural macrophages when compared to control macrophages (A). In addition to this damage, a reduction of mitochondrial respiration and cellular levels of NAD+ was also seen. M40403 (5 – 20 mg kg−1, intraperitoneally) attenuated DNA damage (A) and restored cellular respiration (B) and intra-cellular levels of NAD+ (C). *P<0.01 when compared to control macrophages and oP<0.01 when compared to macrophages taken from M40403-treated rats injected with carrageenan in the presence of M40403.
