Effects of histamine receptor antagonists, indomethacin, and inhibitors of PKA and PKC on VEGF protein expression induced by histamine. The minced granulation tissue (0.4 g) was incubated in 4 ml of EMEM containing 10% (v v−1) calf serum at 37°C for 3 h. After three washes, the tissue was further incubated at 37°C for 6 h in medium containing histamine (10 μM) in the presence or absence of each histamine receptor antagonist or indomethacin (a), and Rp-cAMP, H-89, Ro 31-8415, calphostin C or genistein (b) at the concentrations indicated. VEGF protein in the conditioned medium was detected by immunoblotting and analysed densitometrically. Representative immunoblots are shown at the top. The mean VEGF protein level in the conditioned medium in the control group is set to 1.0. Values are the means from four samples with s.e.mean shown by vertical bars.
