IBMX, Forskolin and IBMX, or Piclamilast inhibit chondrocyte iNOS expression induced by IL-1β. Chondrocytes were exposed to vehicle, 200 pg ml⁻¹ IL-1β or 200 pg ml⁻¹ IL-1β in the presence of 1 mM IBMX, 1 mM IBMX and 10 μM Forskolin or 1 μM Piclamilast over 12 h. Cellular extracts were prepared and immunoblotting was performed with a rabbit polyclonal iNOS antibody as described in the Methods section. Each lane was loaded with 20 μg protein.
