Effect of protein synthesis inhibitor cycloheximide (20 μg ml−1) on (A) time-course of secretin-stimulated (100 nM) cyclic AMP accumulation and (B) cyclic AMP accumulation stimulated by secretin (100 nM), forskolin (10 μM), NECA (10 μM) or iloprost (1 μM). Cells were treated with cycloheximide for 30 min prior to and then during agonist stimulation for the time periods specified. All data are mean±s.e.mean (bars) from at least three independent experiments, each performed in quadruplicate. (A) Over time, secretin-stimulated cyclic AMP accumulation was significantly increased in the presence of cycloheximide when compared to control (P=0.0136, two-way ANOVA).
