Inhibition of Akt does not affect LPS-induced TNF-α release from macrophages. RAW 264.7 cells were transfected with a dominant negative form of Akt (K179M) or a plasmid expressing β-galactosidase as control. After 24 h cells were challenged with LPS (10 ng ml⁻¹; 24 h). Culture supernatants were collected and TNF-α levels determined by ELISA. The inset confirms expression of the dominant negative Akt.
