Dose-response effects of M2 MOs on zebrafish embryo basal and CCh-induced heart rate changes. Each one-cell stage embryo was injected with different doses (0.08, 0.8, 2.4 and 8 ng per embryo) of M2 MO or four mismatch base M2 MO (M2 4 mp MO), and then the embryos were incubated at 28.5°C until 72 h.p.f. for the experiments. (a) The basal heart rate of each 72 h.p.f. embryo was measured at room temperature. (b) The 72 h.p.f. embryo was injected with 10−4 M CCh at room temperature. The ratio of heart rate of CCh-induced bradycardia is defined as in Figure 6 legend. The data represent the mean and s.e.mean from three independent experiments, each with 8–10 individuals. A differences in the M2 MO compared to the M2 4 mp MO control was tested with the Student's t-test and considered significant with P<0.05 (*) or P<0.01 (**).
