Specific binding characteristics of [³H]8-OH-DPAT in peritoneal macrophages. The inset shows a Scatchard plot derived from the specific binding data. The mean binding curves and Scatchard transformations are shown in the figure. The maximum binding capacity (B_max, fmol mg⁻¹) of zymosan- and latex-stimulated macrophages was 1.58 and 1.38, respectively. The affinity constant (K_d, nM) for [³H]8-OH-DPAT was 1.7±0.14 and 1.5±0.08 when determined for zymosan- and latex-stimulated macrophages, respectively. B=bound, F=free. Each sample was run in triplicate. No specific binding was detected in nonstimulated cells.
