Distribution of bodipy-[Leu31, Pro34]NPY after pulse-chase labeling in Y1-transfected HEK293 cells. Cells untreated (left) and treated (right) with monensin were pulse labeled for 40 min with 2 nM bodipy-[Leu31, Pro34]NPY and chased for 0, 10 and 60 min at 37°C in the absence or presence of monensin. In the absence of monensin, there is a progressive disappearance of the fluorescent ligand from intracellular stores, whereas in the presence of the drug, the ligand remains trapped in endosomes due to their inability to dissociate from the receptor.
