Effect of ET-1 on TNFα-induced iNOS expression in differentiated 3T3-F442A adipocytes. Differentiated 3T3-F442A adipocytes were treated for 20 h with TNFα in the presence or absence of increasing concentrations of ET-1. Nitrite levels were determined in the cell supernatant and protein extracts were analysed by Western blot using specific NOS antibodies. A representative autoradiograph taken from three independent experiments is shown. Results are expressed as mean±s.e.m. from three independent experiments. * indicates a statistically significant difference induced by ET-1 when compared to TNFα alone (one-way ANOVA followed by a Dunnett's post hoc test for multiple comparison, P<0.05). A representative autoradiography of three independent experiments is shown. A protein extract from murine macrophages (RAW 264.7) treated with a mixture of IFNγ (10 ng ml⁻¹) plus LPS (1 μg ml⁻¹) was used as a positive control for iNOS.
