SP600125 inhibits JNK activity induced by TNFα (10 ng ml⁻¹) or IL-1β (10 ng ml⁻¹) in human airway smooth muscle cells. Phosphorylation of c-Jun was detected by Western blotting using an antiphospho-specific c-Jun antibody. The blot shown in the upper panel was stripped and reprobed using a phosphorylation state-independent JNK antibody to show the amount of JNK loaded. A representative example of three identical experiments is shown. In the lower panel, densitometric analysis of phospho-c-jun expression, normalized by total JNK expression, is shown. Data are expressed as mean±s.e. of the mean (s.e.m.). *P<0.05 compared to cells treated with TNFα or IL-1β in the absence of SP600125.
