Alkylation of cysteine thiols with N-ethylmaleimide (NEM) prevented the excitatory action of NOCys. (a) Application of NEM (1 mM) to the cytosolic surface irreversibly decreased BKCa channel activity at 20 mV. The subsequent application of NOCys (10 μM) had little effect on BKCa channel activity. (b) comparison of BKCa openings evoked by ramped depolarizations (three ramp depolarizations between −100 and +100 mV every 10 s) in the absence (bi) and 5 min (bii) after the exposure to NEM (1 mM), holding potential 0 mV.
