Effects of PTX on Ca²⁺ mobilisation

Freshly isolated PMNL (1 × 10⁷ ml⁻¹ PGC buffer) were loaded with 2 μM Fura-2 for 30 min at 37°C. Then, cells were preincubated with or without 2 μg ml⁻¹ PTX for 60 min at 37°C. After addition of the indicated stimuli, the fluorescence was measured and intracellular free Ca²⁺ was calculated as described. Values (mean±s.e., n=4) are given as the percentage of Ca²⁺ mobilisation of PTX-treated cells versus control cells that received no PTX.
