JNK activation alone has no effect on cell proliferation. A, DLD-1 cells were treated with either AdGFP or AdMKK7DN at the indicated multiplicity of infection (MOI) for 24 h, and cell lysates were subjected to western blot analysis for the indicated proteins. Lysate from cells treated phosphate-buffered saline (PBS) was used as a control. B, DLD-1 cells were treated with AdGFP or AdMKK7DN at the indicated MOI for 24 or 48 h. The effect of JNK activation on cell growth inhibition was determined by XTT assay. Data represent mean ± SD of three independent experiments. C, DLD-1 cells were pretreated with AdGFP or AdMKK7DN at 2000 MOI for 24 h and then treated with 3 μM DBPT for additional 24 and 48 h. The apoptotic ratio of stained cells with PI was determined by flow cytometry. Data represent mean ± SD from three independent experiments performed in triplicate.
