The RANKL proximal 2-kb promoter is unresponsive to PTH. UAMS-32P cells were stably transfected with luciferase reporter constructs containing either the region from −2014 to +27 of the murine RANKL promoter (RANKL 2kb) or the region from −1379 to +45 of the murine IL-6 promoter (IL-6 1.4kb). Pools of transfected cells were treated with vehicle (veh; white bars), 10−7 M PTH (black bars), or 1.5 mM db-cAMP (striped bars) for the indicated times before determination of relative light units (RLU). Each value represents the mean ± SD of triplicate wells.
