Id-specific CD4⁺ T cells are activated blasts in small MOPC315 tumors in TCR-TG mice. (A) Single tumor cell suspensions were triple-stained with anti-CD4 and anti-CD8 mAbs, and either GB113 or F23.1 mAbs. A combined CD4⁺ and forward/side scatter gate was used, and cells of various sizes (R1, R2, and R3) were analyzed for TCR composition with TCR-TG clonotype-specific (α_Tβ_T) GB113 and β_T (Vβ8)-specific F23.1 mAbs. (B) Tumor cells were triple-stained, and expression of CD25 and CD69 on gated Id-specific CD4⁺ cells is shown, together with isotype-matched control mAbs (shaded areas).
