AS602868 dose-dependently inhibits RANTES expression in kidney cell cultures. Single-cell suspensions obtained after enzymatic digestion of age-matched WT kidney (A) and Tg2 transgenic mice (B) were incubated with increasing doses of AS602868 and 30 minutes later, TNF-α (600U/well) or medium alone was added, the cell supernatants then harvested after 48 hours and analyzed for RANTES expression by ELISA. Each condition was analyzed in triplicate. Data are means ± SD. Experiment was repeated twice (WT, n = 4; CD4C/HIV1 Tg stage Tg2, n = 4), (*, P < 0.05; **, P < 0.01; ***, P < 0.005 versus control without inhibitor).
