Oligonucleotides lacking a 3′-hydroxyl group can be designed to target a specific open complex and inhibit in vitro transcription. The representation of RNAP is not meant to suggest that the enzyme is a single subunit or that it spans the promoter DNA over the sequences as drawn. By convention, the transcription start site is designated +1.
