In vitro analysis of the effect of mutation of the uORF1 termination codon. All transcripts contain the (mutated) Cx41 5′ UTR, the GFP ORF, the X.laevis β-globin 3′ UTR and a tail of 30 A and 30 C nucleotides. Transcripts (Fig. 1C and Table 1) were translated in a rabbit reticulocyte lysate in the presence of [³⁵S]methionine, the resulting proteins were separated on a 15% 30:0.18 acrylamide/bisacrylamide gel, and visualised by autoradiography. Migration of GFP and elongated GFP (#1 GFP) is indicated on the right.
