Cytokine secretion by cultured DC incubated with OVA-BiP hybrid peptide/hsp70 or hsp70. Bone marrow-derived DC from C57BL/6 mice (a–d) or C57BL/10ScN mice (e and f) were cultured for 6 days as described in Materials and Methods and then incubated for 24 h with hsp70 plus or minus OVA-BiP hybrid peptide, with or without polymyxin B, or with tissue culture medium alone. Secretion of IL-1β, IL-6, IL-12, or TNF-α was measured. Hsp70 complexed with OVA-BiP hybrid peptide gave the same results as with hsp70 alone. OVA-BiP hybrid peptide alone induced no significant secretion of cytokines (data not shown).
