Deltex1, Deltex2, and Deltex4 are expressed in T cells, but only Deltex1 is induced by Notch signaling in T cells. (A) Expression of Deltex family members in sorted DN (CD4 CD8), DP (CD4+ CD8+), and SP (CD4+ and CD8+) thymocytes by TaqMan RT-PCR. Relative expression is shown for cDNAs normalized to the value for HPRT. (B) Expression of Hes1, Deltex1, Deltex2, and Deltex4 in 1010 expressing empty vector or the intracellular domain of Notch1 or Notch3. The relative expression of each gene was normalized to that for HPRT. (C) All three Deltex family members can inhibit Notch signals. Cos-7 cells expressing a CBF1/RBPJκ-responsive element (8xCBF1) linked to GFP were transiently transfected with Myc-tagged Notch-IC alone or cotransfected with Myc-Notch-IC and Flag-tagged Deltex1, Deltex2, or Deltex4, and the total percent GFP+ cells was determined by FACS analysis. Average inhibition of GFP expression in cells cotransfected with Deltex1, Deltex2, or Deltex4 compared to that in cells transfected with Notch-IC alone is shown. Error bars show average data from four independent transfections using Notch1-IC, Notch2-IC, or Notch3-IC.
