Nucleocapsids and glycoproteins colocalize in extracellular purified virions. HSV-1 F-VP26/GFP virions were purified from Vero cell culture supernatants, immobilized on collagen-coated glass coverslips, and stained with mouse anti-gE/gI-specific MAb (A to C) or rabbit polyclonal gD-specific antibodies (D to F) followed by FITC-conjugated anti-mouse IgG or Texas red-conjugated anti-rabbit IgG. Filled arrows in panels A to C represent a particle that contained glycoproteins and little or no capsid signal. Empty arrowheads in panels D to F represent a particle with high levels of capsid fluorescence compared with anti-gD staining.
