The Golgi apparatus and microtubules are required for the transport of cholesterol and sphingomyelin to the chlamydial inclusion. Infected HeLa cells were incubated with [14C]squalene, [3H]cholesterol oleate/LDL, or [14C]ceramide in the presence of BFA or nocodazole to inhibit Golgi and microtubule functions, respectively. Radioactive cholesterol or sphingomyelin associated with EBs were monitored. Data are expressed as mean ± SD. (Lower) A representative immunoblot against the chlamydial major outer membrane protein (MOMP) from density gradient-purified EBs to show equivalence in the samples used in the analysis of incorporated isotope.
