fNMHCB retains PBX1A in the cytoplasm of Drosophila S2 cells. Drosophila S2 cells were transfected with insect expression vectors for HA-PBX1A, MEIS1A, GAL-DBD, and GAL-fNM individually or in combination. Immunostaining was performed with anti-HA and anti-MEIS1 antibodies. DAPI staining reveals nuclei. (A and B) PBX1A is located in the cytoplasm of S2 cells. (C to E) Expression of MEIS1A induces nuclear translocation of PBX1A. (F to H) GAL-DBD has no effect on the subcellular localization of either PBX1A or MEIS1A, whereas GAL-fNM opposes the action of MEIS1A, inducing cytoplasmic retention of PBX1A, in a dose-dependent manner. (I to K) Equal amounts of expression vectors for GAL-fNM and MEIS1A were transfected. (L to N) The ratio of transfected GAL-fNM to MEIS1A expression vectors was 5:1.
