Growth of the MLM369 (dnaB^–/ban⁺) strain at 30 and 42°C. Single colonies of MLM369 (dnaB^–/ban⁺, open circles) or MLM370 (dnaB⁺/ban^–, filled circles) were picked and grown overnight at 30°C on LB medium with kanamycin. Cultures were then diluted in the same medium to an A₆₀₀ of 0.02 to 0.04 and were split into two cultures; one was grown at 30°C (A) and the other at 42°C (B), both with the same shaking and aeration conditions. Samples were removed every hour to measure optical density at a wavelength of 600 nm. The graphics show a typical pattern, which was reproduced in three independent experiments.
