GH stimulates the expression of Foxa-2 in primary preadipocytes of lean and obese mice. Analysis of gene expression in the stromal vascular fraction (containing preadipocytes) of wild-type (lean, C57/B6) and ob/ob mice that were cultured in the absence (0) or presence of 500 ng/ml growth hormone for 2, 8, and 24 hours. Foxa-2 and Pref-1 expression levels were increased (2.5 and 3.5, respectively; P ≤ 0.05) in 2-, 8-, and 24-hour time points compared with untreated cells. Socs-1 expression was increased at 8 and 24 hours (1.9 ± 0.2; P < 0.05). Expression levels in ob/ob preadipocytes were significantly increased after 8- and 24-hour stimulation compared with unstimulated cells (Foxa-2: 2.6 ± 0.2 and 4.8 ± 0.4, P ≤ 0.05 and P ≤ 0.01, respectively; and Pref-1: 2.4 ± 0.3 and 2.3 ± 0.1, respectively, P ≤ 0.05). Socs-1 was increased at 8 and 24 hours (1.9 ± 0.1 and 2.1 ± 0.3, respectively, P < 0.05). No significant changes in expression levels of Foxc-2 were detected.
