Dry mature pea seeds were imbibed for 18 h at room temperature in circulating water and then planted in vermiculite (d 0). Growth was conducted at 22°C with a photoperiod of 12 h. Folate was determined using a microbiological assay with Lactobacillus casei. Values are means ± se of three separate experiments, each performed in triplicate. The amount of the HPPK-DHPS protein was analyzed by western blotting using 60 μg of soluble proteins for each sample. Quantification of the mRNA coding HPPK-DHPS was not realized because the extraction of total RNA from dry seeds and cotyledons using the RNeasy Plant Mini Kit (Qiagen USA) results in low amounts of RNA, which are not suitable for RT-PCR experiments in our standard conditions.
