Insertion of random hydrophilic amino acid stretches into Δ20 rescues expression and activity. E. coli T184 harboring a plasmid encoding the Δ20 mutant with a given hydrophilic insertion in the middle cytoplasmic loop were assayed for lactose transport (Upper) and membrane expression of the permease (Lower) as described in the text.
