Insertion of random hydrophilic or hydrophobic amino acid stretches into the Δ12 or Δ5 mutant. E. coli T184 harboring a plasmid encoding either Δ12 (A) or Δ5 (B) with a given hydrophilic or hydrophobic insertion in the middle cytoplasmic loop were assayed for lactose transport (Upper) and membrane expression of the permease (Lower) as described in the text.
