FIG. 2. — Characterization of polyclonal rabbit anti-CXCL16 antibodies. A B300.19 cell line was transfected with a plasmid encoding CXCR6 or an empty plasmid as control. These cell lines were evaluated for their ability to migrate toward soluble CXCL16. (A) A range of CXCL16 concentrations were used, and transfected CXCR6⁺ cells displayed a classic bell-shape chemotaxis profile with peak migration occurring at 100 ng of CXCL16/ml, while no migration was observed for the control. The ability of polyclonal rabbit anti-CXCL16 antibodies to block CXCL16-induced migration was tested in a chemotaxis assay. (B) Complete inhibition of CXCR6⁺ cell migration toward CXCL16 was obtained with concentrations of anti-CXCL16 of ≥30 μg/ml. These data are representative of three experiments performed with similar results.
