Hyal1 mRNA is expressed in metastatic and non-metastatic prostate carcinoma cells, but only metastatic cells express HAS2 and HAS3. Equivalent amounts of input poly(A)+ RNA from prostate carcinoma cell lines (LNCaP, 22Rv1, PC3, and PC3M-LN4) or a normal prostate mRNA library (N.Pr.) were reverse-transcribed, and target messages were amplified using oligonucleotide primers specific for Hyal1, HAS2, HAS3, or glyceraldehyde-3-phosphate dehydrogenase (housekeeping control) as described previously and in Materials and Methods. An aliquot of 20 μl from each PCR was electrophoresed on a 2% agarose gel and stained with ethidium bromide for digital imaging.
