The dependence on pH of the rate constant for 18O exchange RH2O/[E] catalyzed by H64W HCA II and measured in the absence of buffer (▪), and in the presence of 75 mM 4-MI (○). Data representing the difference between these two sets are shown (Δ). Data were obtained at 25°C using solutions containing 25 mM of all species of CO2. Ionic strength was maintained at a minimum of 0.2 M by addition of sodium sulfate. The solid line for the difference data was obtained by a fit to Eq. 8 resulting in Inline graphic for the proton donor(s) was 8.3 ± 0.1 and pKa for the zinc-bound water was 6.7 ± 0.1.
