ATP hydrolysis reactions were as described under ‘Materials and Methods’ using 77 nM BACH1-WT or BACH1-M299I and 82 nM of the forked duplex effector (Table 2, DNA molecule 1) for the kcat determination. For Keff determination, 50 nM BACH1-WT or BACH1-M299I was used with the forked duplex DNA effector concentrations ranging from 0 to 75 nM.
