The mutation S967A accelerates ASK1-induced cell death. HeLa cells were cotransfected with expression vectors for HA-tagged ASK1^wt, ASK1^S967A, or ASK1^K709R and a vector for β-gal. At the indicated times, attached cells were harvested and assayed for β-gal activity. This activity reflects the viability of transfected cells only, because dead cells will detach from the culture dish. The control sample was transfected with an amount of pcDNA3 equal to the amount of ASK1 DNA. Results shown are mean ± standard error (n = 3) and are representative of three independent experiments.
