Fluorescent immunohistochemical analysis of PBX1 siRNA-transfected MCF7 cells. Immunofluorescence confocal microscopy was conducted for PBX1 (red) and VCP (green). Merged images are shown. PBX1 and VCP expression in PBX1 siRNA clone was weaker than those in negative siRNA clone and MCF7 cells. Control analyses with nonimmune rabbit or mouse serum as primary antibodies instead of anti-PBX1 and anti-VCP antibodies showed the fluorescence for the specific antibody, but not for the nonimmunized serum, indicating the specificity of primary antibodies and absence of bleed-through of the fluorophores.
