Optimisation of virus concentration for gene transfer. Corneal samples were transduced with up to 1.5 × 108 pfu AdCL per triplicate specimen. Assays were repeated twice with consistent results. Mean values (SD) from one assay are shown. (A) Catalase quantification by ELISA. The amount of catalase detected by ELISA increased with the amount of AdCL virus applied, reaching 730 mU after incubation with 1.5 × 108 pfu. (B) Catalase quantification by H2O2 activity assay. Corneal samples were infected with up to 1.5 × 108 pfu AdCL. The amount of catalase enzyme detected by ELISA increased with the amount of AdCL virus applied, reaching 795 mU after incubation with 1.5 × 108 pfu. The curve reached a plateau at approximately 5 × 107 pfu AdCL.
