Luteolin inhibits LPS-dependent NF-κB transcriptional activity in BMDCs isolated from cis-NF-κBEGFP. Bone marrow-derived dendritic cells (BMDCs) isolated from cis-NF-κBEGFP mice were pretreated with various concentrations of luteolin for 1 hr, and then stimulated with LPS (1 µg/ml) for 18 hr. Cells were lysed using Nonidet P-40 lysis buffer and EGFP levels measured using a Gemini XS microplate reader. The results were normalized to protein extract concentrations. The excitation and emission settings were 488 nm and 509 nm, respectively. Data are expressed as percent changes over control determined as the means of three independent experiments.
