Mutations in Casp8–AR interaction motifs abolish Casp8-mediated inhibition of AR transactivation. (A) Casp8 with mutations in interaction motifs lost its ability to inhibit AR-driven transcription. PC3mm2 cells were transfected with pGL3-4 × ARE-E4-luc, pcDNA-AR, pcDNA-Casp8 (wild-type), or pcDNA-Casp8 (F45A, I174A/I175A, or F295A/I296A) mutants. Transfected cells were grown in the presence of 10 nM R1881 for 48 h and then harvested for luciferase assay (top panel) and RT–PCR. (B) Mutations in interaction motifs in AR abolish Casp8-mediated inhibition of AR transcriptional activity.
