Proviral effects of naturally existing ADAR1 variants. (a) Different ADAR1 variants, including LF150, Dcat, SF110, SF80, and the vector control, were introduced into wild-type (WT) MEFs. Stable cells were infected with the recombinant VSV-EGFP1 (MOI = 10), and infections were compared for different ADAR1 variants as described above. Note: as the basal level of VSV infection was high in PKR−/− MEFs, the differences for these ADAR1 variants were not as significant as in WT MEFs (data not shown). (b) Comparison of the naturally existing ADAR1 variants on PKR activation. 293T cells were transfected with the same set of ADAR1 constructs; p-eIF-2α and total eIF-2α in cell lysates were determined by immunoblotting and quantitated with Scion Image software. The ratios between p-eIF-2α and eIF-2α were calculated for each construct and normalized to the vector (*). All data are shown as mean plus standard deviation (n = 3).
