FoxM1-deficient cells exhibit increased DNA breaks. (A to D) To examine for DNA breaks, wild type (+/+) or FoxM1−/− (−/−) MEFs were immunostained for phosphospecific histone H2AX (γH2AX) foci or for TUNEL foci. FoxM1−/− MEFs exhibited high levels of spontaneous γH2AX foci (A and C) or TUNEL-positive foci (B and D) compared to wild-type MEFs. (E and F) Osteosarcoma U2OS cells depleted in FoxM1 levels by siRNA transfection exhibited high levels of spontaneous γH2AX foci. U2OS cells were transfected with FoxM1 siRNA or control siRNA or received no treatment, as indicated. Cells were then exposed to 5 Gy of IR and immunostained for γH2AX foci at 0 h (E) or 24 h (F) post-IR. The asterisks indicate statistically significant changes: **, P ≤ 0.01; ***, P ≤ 0.001.
