Monocyte production of intracellular TNF-α and IL-12 PBMCs from 12 healthy non-pregnant and 12 normal pregnant women were incubated for 6 h with endotoxin (0·04–40 ng/ml), IFN-γ (5·5 ng/ml) and monensin (2·4 µm), or with monensin only. The cells were double-labelled for CD14 and intracellular TNF-α or IL-12, and 60 000 events were analysed by flow cytometry. The percentage of CD14+ cells which were TNF-α positive (a) and the brightness of TNF-α positive monocytes (b) were calculated. Significant differences are shown between non-pregnant and pregnant groups (*P < 0·05).Monocyte production of IL-12 was analysed in the same way (c, d).
